The Spitfires are now in the hat to face the giants of the Premier League - including Saints - after second half goals from James Constable and Josh Payne.
Constable finished at the near post just minutes after Ben Billingham had almost given Stourbridge the lead.
And Josh Payne then made sure of the result as he finished off Ben Strevens' low cross.
There was still an opportunity for the home side to halve the deficit but Karl Hawley's penalty was saved by Eastleigh keeper Ross Flitney.
Eastleigh are now in the third round for the first time, and will find out their opponents on Monday.
