James Taylor hit a 77-ball 115* as Nottinghamshire made 277-4 after Michael Lumb was lbw for 0 on his return to Hampshire.
But half-centuries from Jimmy Adams, Michael Carberry and Sean Ervine - and a brave 48 from Dimi Mascarenhas - took the game to the final over.
Hampshire needed 15 from the last six balls, but Darren Pattinson held his nerve as the Royals suffered their first CB40 defeat of the season, by 12 runs.
